Aging is associated with reduced anabolic hormone activity including growth hormone, thyroid hormone and sex steroid axis activity with increased abdominal fat (insulin resistance). Lean mass contains the body’s protein content. Every protein molecule has a role in maintaining body homeostasis. Loss of any body protein is deleterious. “Inflammaging”
In the elderly, hyperstimulation of the immune system results in a chronic, low-grade state of inflammation. Inflammaging is macrophage centered, involves several tissues and organs, including the gut microbiota, and is characterized by a complex balance between pro- and anti-inflammatory responses. The major source of inflammatory stimuli is represented by endogenous/self, misplaced, or altered molecules resulting from damaged and/or dead cells and organelles (cell debris), recognized by receptors of the innate immune system. While their production is physiological and increases with age, their disposal by the proteasome via autophagy and/or mitophagy progressively declines.
Loss of control of the ‘autoreactive/autoimmune’ process fuels the onset or progression of chronic diseases that can accelerate and propagate the aging process locally and systemically. Consequently, inflammaging can be considered a major target for antiaging strategies, and includes decreased microbiota diversity and stability, along with misfolded proteins spreading along the gut-brain axis.
Oxidative Stress and Mitochondrial Dysfunction
Unmanaged oxidative stress and impaired cellular stress responses are also central to aging. Current research reveals that rather than applying high dose anti-oxidants, the emphasis should be on consuming compounds that activate endogenous defense pathways.
